Mekhi Phifer was born in Harlem, New York City. He is the son of Rhoda Phifer, a high school teacher. He was raised in a single-parent household with his mother.
In 1995, he started his acting career after auditioning at an open casting call for director Spike Lee's 'Clockers'. He was selected for the lead role. His role was as narcotics dealer involved in a murder cover-up. He continued with the same role in the comedy film 'High School High' and 'I Still Know What You Did Last Summer'.
He became famous for his role as Gregory Pratt on the medical drama 'ER' in 2002. He was nominated twice for an NAACP Award for 'ER'. He left this show
in September 2008.
Few of Phifer's movies include 'The Tuskegee Airmen' (1995), 'HBO's Subway Stories: Tales from the Underground' (1997), and 'Brian's Song' (2001).

In 2002, Phifer starred as Future, opposite Eminem, Brittany Murphy and Kim Basinger in Curtis Hanson's critically acclaimed film, '8 Mile', and in 2002, Miramax feature 'Paid in Full' (2002), directed by Charles Stone III. In June, he was honored by the American Black Film Festival, receiving the organization's 'Rising Star' Award.
Phifer married Malinda Williams, an actress in 1999 and they have a son named 'Omikaye'. The couple divorced in 2003. Phifer has another son, Mekhi Thira Phifer Jr., who was born to his Laotian mistress Oni Souratha in Los Angeles on 30 October 2007.
