Lane was born in New York City. She is the daughter of Colleen Farrington and Burton Eugene Lane. Her parents were separated when she was a few months old. She lived with her mother till she was 6 and then moved with her father.
When she was 15 she ran to Los Angeles and came back and moved to a family friend. She paid them rent and in 1981 she enrolled in high school.
She began acting professionally at the age of six at the La MaMa Experimental Theater Club in New York. When she was 12 years old she had a role in, The Cherry Orchard. She turned down a role in Runaways on Broadway to make her feature film named A Little Romance

She was featured on the cover of Time, next year at the age of 14. She appeared in many more films with famous stars. She turned down two films to appear in The Cotton Club which was a failure and her career languished so she moved with her mother in Georgia.
She returned to the film industry to make a few more movies. In 1989 she appeared in Lonesome Dove and gained a lot of popularity. She was nominated for an Emmy Award for her role in the television series. Her performance was well received and she went to the Cannes Film Festival.
Lane appeared in another movie in 1999. She won an Independent Spirit Award nomination for Best Female Lead. In 2002, Lane starred in Unfaithful, in which she played the role of a housewife. Lane, in 2008, reunited with Richard Gere for the movie Nights in Rodanthe. In 2008, Lane expressed frustration with being typecast and is gunning for something that's not so sympathetic. In 2009, it was announced that Lane will star in Secretariat.
Lane married Lambert in 1988 in Santa Fe, New Mexico. They had a daughter, Eleanor but were divorced in 1994. Later she was engaged to Josh Brolin in July 2003 and they were married in 2004.
